- The real Jonathan Larson received three posthumous Tony Awards and a posthumous Pulitzer Prize for the rock musical "Rent".
- The prototype for Michael, who has HIV, played by Robin de Jesus, was a man named Matthew O'Grady, who was, fortunately, still alive in 2021.
- Andrew Garfield had to learn how to sing for the role of Jonathan Larson. It took the actor a year to do so.
- After casting Andrew Garfield as the lead, but remaining unaware of whether he could sing, director Lin-Manuel Miranda asked a mutual friend about it. The friend replied that he sings beautifully and has an "angelic voice." Once Miranda left, this mutual friend called Garfield and asked if he actually knew how to sing.
- Originally, Jonathan Larson (1960–1996) envisioned 'Tick, Tick... BOOM!' as a one-man show. In 2001, playwright David Auburn reworked it into a piece for three actors. The film features even more characters.
- The scenes in which Jonathan Larson performs 'Tick, Tick... BOOM!' were filmed at the New York Theatre Workshop, where Larson's smash-hit musical 'Rent' officially premiered in 1996.
- The film several times mentions composer, lyricist, and playwright Stephen Sondheim (1930–2021), the author of many musicals, whom Jonathan Larson deeply revered and considered his mentor. Sondheim provided invaluable assistance to Lin-Manuel Miranda during the making of the film, but unfortunately passed away on November 26, just a few days before the premiere.
- The character of Michael, who is HIV-positive and played by Robin de Jesús, was based on a certain Matthew O'Grady, who, fortunately, was still alive in 2021.
